EGCG also inhibits activation of the EGFR in carcinoma cells, down-regulation of ER-α36 expression was in accordance with down-regulation of EGFR, which further verified a loop between ER-α36 and EGFR, GEN and EGCG reduced BRCA1 CpG methylation and cell proliferation associated with increased p53, The expression of FAS was also suppressed by the tea polyphenol (-)-epigallocatechin 3-gallate (EGCG) at both protein and mRNA levels that may lead to the inhibition of cell lipogenesis and proliferation. In all, EGCG is the first identified GOLM1 downregulation natural product. Western blot analysis revealed EGCG induced a concentration-dependent decrease in ERα protein levels, with a 56% reduction occurring with 60 µM EGCG when compared to controls. EGCG NPs modulate the expressions of several key regulatory proteins in PI3K-Akt pathway such as up-regulation of PTEN, p21 and Bax, and down-regulation of p-PDK1, p-AKT, CyclinD1 and Bcl-2 (MCF-7) |
